  | In culture, these cells mimic the multistep process of chondrocyte differentiation. After plating, they proliferate until confluency, at which point they undergo growth arrest following contact inhibition. After 4 days, these cells undergo a phenotypic change characterized by the secretion of a variety of ECM molecules, such as type II collagen, aggrecan, and link protein, and by the formation of cartilaginous nodules on the culture plate (12, 18, 19).

  | A multipotent factor-dependent cell line, FDCP-mix. These cells possess a nonleukemic phenotype, have a normal karyotype, are factor dependent, and can be directed to undergo multilineage development in response to cytokines (77). By changing growth conditions (serum and cytokines), it is possible to grow this cell line under self-renewing conditions, such that they retain their multipotent state; alternatively, they may be directed to differentiate to form erythroid or myeloid cells. 16648489(C)

 |
  | HC11 mouse mammary epithelial cells have been widely used as an in vitro model of mammary gland epithelial cell differentiation. The HC11 cell line preserves important features of mammary epithelial cell lactogenic differentia- tion; it was clonally derived from the COMMA-1D cells, a line immortalized from mammary tissue of a pregnant BALB/c mouse [1,2]. The HC11 cells are non-tumorigenic, display a normal epithelial phenotype, and the injection of HC11 cells into the cleared fat pad of BALB/c mice exhibited normal ductal and alveolar-like structures [1,3]. HC11 mammary epithelial cell lactogenic differentiation can be initiated in culture following the growth to conflu- ence and deposition of extracellular matrix in the presence of epidermal growth factor (EGF), subsequent removal of EGF from the culture and the addition of lactogenic hormone mix, DIP (dexamethasone, insulin, and prolactin); upon differentiation HC11 cells express specific milk pro- teins including β-casein [1]. Moreover, during lactogenic differentiation in culture the HC11 cells undergo pheno- typic transformation to "mammospheres", enlarged domed structures with a lumen [4-6].

  | Adapted to CD 293 Medium (Cat. No. 11913-019).293-F and 293-H Cells are clonal isolates, derived from transformed embryonal human kidney cells (1,2). The 293-F strain is a fast-growing variant of the 293 cell line. The 293-H strain is a variant of the 293 cell line that demonstrates better adherence in monolayer culture and ease of use for plaque assays and other anchorage dependent applications. The 293 parental cell line was selected for growth and transfection efficiency (2). Both 293-F and 293-H cell lines can beadapted to serum-free suspension culture in 293 SFM II or CD 293 Medium supplemented with L-glutamine for recombinant protein production and adenovirus propagation.

  | Primary fibroblast cells from a classical Menkes disease patient were immortalized by the SV40 gene transfer to derive Me32aT22/2L (43) and designated MNK(Del). The 4.6-kb cDNA encoding the human MNK protein was cloned into a mammalian expression vector and transfected into Me32aT22/2L, and MNK-expressing clone A12-H9 was designated MNK(++) (43). The empty mammalian expression vector, pCMB77, was transfected alone into Me32aT22/2L to derive Me32aT22/2L(pCMB77) and designated MNK(v/o) (43).

The 3T3-L1 cells were then incubated with DMEM containing 100 U of penicillin per ml, 100 ug of streptomycin per ml, and 10% fetal bovine serum for 8 to 40 days to maintain the adipocyte phenotype.

  | Derived from the peripheral blood of a 1 year old male with acute monocytic leukaemia. THP-1 cells have Fc and C3b receptors and lack surface and cytoplasmic immunoglobulins. These cells also stain positive for alpha-naphthyl butyrate esterase produce lysozymes and are phagocytic (both latex beads and sensitised erythrocytes). THP-1 cells can also restore the response of purified T lymphocytes to Con A, show increased CO2 production on phagocytosis and can be differentiated into macrophage-like cells using for example DMSO.
